The rule of thumb is to try to match the flywheel inertia to the majority of your events.

• Flat and fast road riding = large gearing and faster flywheel inertia

• Hilly and off-road = small gearing and slower flywheel inertia

Big or small chainring on kickr?

Gearing does have an impact. It is essentially affecting the flywheel speed and inertia, which impacts how it loads your body and muscles. Here is a video showing some of the differences.
